What about moving another comp account to this OU and testing?

The same GPO applies without any issue to all the other OUs it is linked to. This particular OU is a test one so has Block Inheritance on it, but this GPO is applied directly to the OU. There are no WMI filters and no security filters applied, I have tried restarting the server that is in the OU but the problem remains the same.
