Nuke autosaves every X number of seconds, where X is what you have defined
in the AutoSaveTime knob in the Nuke preferences. With default
configuration, the method it uses to autosave is to place a
"<scriptname>.nk.autosave" file beside your project file. This should be a
viewable non-hidden file on all operating systems.

A quick test reveals that when you quit Nuke with unsaved changes, and say
"No" to the save changes dialog box, the autosave file is deleted. This
makes sense, since if you wanted to not save your changes, and it didn't
delete the autosave file, Nuke would prompt you to restore the autosave file
when you started started it up again.

Therefore, as far as I know, there is no way to restore changes to a script
which you specified to not save changes to.

The *.autosave file is just a copy of your nuke script though, so for
example if Nuke crashes, and you want to version up your script, you can
just rename the <scriptname>_v01.nk.autosave file to <scriptname>_v02.nk,
and it should work fine.

In response to Randy Little's clarification, it does look like the autosave
file is preserved in place if you say no to recover autosave. However, when
you save the script again with new changes, the autosave file appears to be
destroyed.

There's always the option of data recovery solutions, but that might be a
little extreme for the circumstances.
