Nuke's rotopaint nodes does have strait line functionality built in.... however 
it doesn't work very well. If you need precision then use shape/stroke point 
methods that everyone has been suggesting. The other way goes as follows:

1) start drawing a stroke
2) without lifting your pen up hold down the ctrl key
3) you will see the cross hair show up in the middle of the brush
4)now you can lift up the ctrl key and the brush
5) move to the new locaton
6) start painting, do not just click

It will draw a strait line between the two points. It is kinda complicated, and 
sorta touchy. It doesn't always work how you would expect it to.
