Are any parts of that path NFS-mounted? Is SELinux or similar enabled? Does the path even exist?
Actually, I think you're likely seeing the fact that nxlog runs as a particular service account (eg. nxlog). It is that user that will need to be able to read (and write, I think) to that file. Bit odd to see /usr/local/var/... If building from source (the packages are good to use if you can) then make sure you configure it to have the state-dir as /var/...
