Been there, seen it, and it's not worth the trouble.

Shimano STI levers, especially the newer units (e.g. Ultegra
6500-6501-6510 units) are designed not to be taken apart. It's
definitely not Campy - if the lever dies, the whole thing dies and you
might as well buy a whole new lever. Having said that, you can replace
the blade portion of the lever, or the part that has the inner and outer
levers (if Ultegra, you'll be looking around $300 with tax, that's
almost the same cost as a brand new lever!) but it's a little tricky.
Shimano even recommends a special tool for doing it (and it's no doubt
pretty expensive...) As for opening up the internals of the blade...
it's like you're a baby trying to reassemble a swiss watch blindfolded.
Too many small parts, and your fingers never seem to be small enough.

My best advice would be to check out these links for exploded views of a
typical STI lever (more diagrams are available from this site:

One last bit of advice: don't spray WD-40 into the internals if STI
shifting starts getting mushy or uneven. The stuff will eat up some of
the more delicate plastic components.
