To create a macro, select the modules you want to go into the macro (which cannot include interactors, Image, Display or Colormap - these will have to be inputs to your macro). Then under the Edit menu, theres a Create Macro ... button. Press it, enter a macro name and the name of a file in which to store it, and OK. When you run OpenDX again, you can File -> Load Macro... to load it, or add the directory in which you placed it to your DXMODULES environment variable, or add "-modules dirname" to the command line with which you start OpenDX.
