Do you mean programmatically or by drawing a line?

For the first option: each geometry has a distanceTo function which you can 
use. If you need to measure on a sphere, check out OpenLayers.Util.distVincenty 
instead.

If you need a control to do the drawing and return the distance, check out 
OpenLayers.Control.Measure:
