Has the library been closed?
If it was opened, but has not been closed (and unloaded?) Hotsync will
fail to replace the library with the above error message.
A sort reset should fix this problem.
If you have to leave your library open most of the time, consider
registering for the Hotsycn start/finish notifications, and unload you
library between them to allow hotsync to replace it.
