One or the other; they are not technically two ethernet cards, but rather two 
connectors on the same ethernet controller.

A PCI ethernet card is a second controller and the Mac will show a second Ethernet 
option in AppleTalk and TCP/IP (if the drivers are installed, of course).

You can configure one, or the other. Mac OS 7-9 do not natively multihome.

You'll need some gateway software, which exists in abundance but is not free. 
IPNetRouter is the most widely used.
