I don't see why not.  However, if you do that, you'll always be left
with a drive with 2 partitions.  If that's OK with you, that's not a
problem.  Otherwise, you'll have to have something big enough to store
what you have in both partitions and performa low level format to
create only a single partition and reload your stuff.
