Luminance smoothing can help smooth out the grainy look you get in
shadow areas of high ISO images or generally underexposed high ISO
images. There seems to be a price in detail, but I've used it on
portraits and the like quite effectively. Color noise reduction can
reduce the muliti-colored mottling that can be seen in some constant
color areas of high ISO and underexposed images. When using either
control, you should be examining various areas of the shot at 200% or
so. This will give you a good look at what you're accomplishing. For
normal images, both can usually be turned all the way off. Sometimes
I'll use a small amount of color noise reduction just to help out in
shadows. Luminance reduction is almost always at zero unless I'm
working with a problem image.
