Here's what I do:

1. Bring in the transparency at full depth.

2. Run levels, tweak until I'm happy.

3. Colour correction, if still needed after levels.

4. Rubber stamp out dust, dirt, marks.

5. Convert to 8-bit.

6. Sharpen, etc.

The extra depth will really help you if you do a lot of contrast
correction to the original.  If the original, on the other hand, is just
right as it is, you can get away with scanning at 8-bit...but why
bother, unless you're in a hurry.
