The relevant value is PERL5DB. It should have been set by pdkdebug for you, but maybe there is some permissions issue if you don't run it elevated.
pdkdebug will set this variable in the registry under the Perl key, but you can also add it manually to the environment yourself. The value should be something like: "{BEGIN {require q<C:\...\bin\lib\PerlDB.pl>}}" without the quotes, and you have to figure out the exact path yourself (it may not even be under the PDK/bin/lib directory in PDK6, I can't remember).
