You should be able to use either one, but quoting may be easier, or get you 
going faster.
Something like:
rsync -aPS foo user@host:”/path/to/dir with spaces/“
should work. Likewise, 
rsync -aPS foo user@host:/path/to/dir\ with\ spaces/
should also work

It gets more complicated if there are escape characters in the directory or 
file name. Hopefully that isn’t happening.
