We in our company had the very fortunate/unfortunate chance (as we're in
the business of making and customizing free software for corporations
and governments)  of exploring at least three viable GUI designers :
wxDesigner, Boa Constructor and wxGlade.

wxDesigner follows a sizer-based layouting scheme for dialog generation,
with the unfortunate side effect of lesser flexibility, while Boa
Constructor makes it a bit difficult inserting foreign classes.
wxGlade... if you're quite comfortable with the Glade approach of
constructing widgets, then this might be a good candidate, although it's
not exactly WYSIWYG. 

So far the best GUI designer we've used (in terms of usability and speed
of getting things done) for wxPython is still the proprietary, non-free
(beer-wise :D) wxDesigner by Rob Roebling, although the free software
Boa Constructor follows. wxGlade is quite good though if you've already
got pre-existing code that you would just want to plug-in. 

Choose your own poison :D
