Do I need to change my /etc/postfix/main.cf to be able to make Spamassassin work? I have installed Spamassassin but do not know how to config.
If you installed spamassassin, it is enabled by default in amavisd.conf.
ALso the system wide config for soft whilelisting/blacklisting is in amavisd.conf.
Note that amavisd uses a system wide spamass config. ( not per user )

No it will not. Either install the Mail::SpamAssassin perl module or use dspam.
My current installation is
Postfix + courier + mysql + amavisd-new + clamav
