Oh yes it will. Mailing lists decay over time as people change their email addresses and forget to update their details with any opt-in lists they've subscribed to. Bounce handling is, therefore, an essential aspect of list administration - you need to be able to update the list regularly to remove inactive addresses from it. Lack of bounce handling is precisely the sort of thing that can get an otherwise entirely reputable list flagged as a spam source.
