There's no reason Postfix won't work with /var/spool/postfix mounted from RAM. The standard filesystem(s) used for ramdisks may not work, you may need to pick a real file system like ext3/4, but if you have something you can treat as a block device there should be no technical issues.

Whether you *should* do something like this is an entirely different matter.
