You need to do two things. First, enable random signatures for the
account. from Setup > Mail Accounts...

In the Default Text Signature, choose Random Text Signature.

Then in Setup > Text Signatures... select a text signature you want to
use in your random list, then click the Include in random list box.

That will do it.
