Sounds like your frontmost document is in bitmap mode.

The Color Settings dialog has "Save" and "Load" buttons. The keyboard
shortcuts dialog has a save icon that looks like a disc. But you don't need
to use either of these, or save your plugins or presets folders. These are
not where any of your settings are saved. (Exception: you may have third
party plugins that have been installed in your plugins folder, and you may
want to save those rather than re-install them after re-installing
Photoshop).

All your settings, presets, etc. are saved in your "Adobe Photoshop CS
Settings" folder, which is saved in a local (per-user) folder and is not
affected by reinstalling the application.

1. The first thing to try is just resetting your preferences. If you don't
want to set your color settings again, save them first (just save them in
the default location and they'll show up in the popup menu in the future).
To reset your preferences, hold down cmd+opt+shift (ctrl+opt+shift  on
Windows) while launching Photoshop and answer Yes when asked if you want to
reset the settings.

2. If you want to reinstall (this is almost never necessary), just put the
adobe photoshop cs folder in the trash (on Mac) or use the uninstall option
on windows, and then re-run the installer. All your settings will be
preserved.
