PyGTK and PyGTK applications should work fine on OS X if you are using the Apple X Server for OS X (they do not run as native Cocoa applications).

For what it's worth, Immunity's product, CANVAS, which is a fairly complex, threaded pyGTK program, works out of the box on OS X in this manner. It looks great! (I've only seen a screenshot - we don't run OS X ourselves, which is part of the coolness factor of it.)
