PyGObject should be good to go, all win32 patches have been accepted. GObject-Introspection still has a couple of patches currently in review and some that need to be reworked (due to an unfortunate behavior of MSYS bash). See https://bugzilla.gnome.org/show_bug.cgi?id=620566 GTK+ 3 at this point will need a bit of work too before it will be as stable and usable as GTK+ 2.
