In mock (at least 5000 lines of code including tests) I target 2.4 -> 3.2+. Admittedly mock does little I/O but does some fairly crazy introspection (and even found bugs in Python 3 because of it). The exception handling is the worst - no compatible syntax between 2.4-5 and Python 3. So you have to use sys.exc_info. Other than that it isn't too hard / bad.
