Something like this...not my code...untested...
im = Image.open("image.jpg")
opacity = .5
if im.mode != 'RGBA':
im = im.convert('RGBA')
else:
im = im.copy()
alpha = im.split()[3]
alpha = ImageEnhance.Brightness(alpha).enhance(opacity)
im.putalpha(alpha)
