There should be no need to manual editing of the file. In the described example, one issue could be the 3.0 API change: QgsColorButtonV2 doesn't exist anymore in 3.0. So one needs to use the proper library of custom widgets (Qt5) to be compatible.
If you compile the UI, it is import to add QGIS to the python path so it can find the custom widgets while compiling.
