You can make QGIS portable by installing with OSGeo4W directly to your USB
stick or on a folder you'll move after (for example into E:\OSGeo4WPortable
or E:\QGISPortable)

Just start the qgis.bat ie your E:\OSGeo4WPortable\bin\qgis.bat (or
E:\QGISPortable\bin\qgis.bat)

That file ( OSGeo4W.bat in fact) takes care of setting environnement
variables from the installed folder.
