This sounds pretty much as a QMail problem, which is not the topic of this 
list...
But, OK, here goes:

Since you don't have to restart qmail in order to run the test, it surely 
is a QMail problem.
And since you probably did a "make setup check" in order for qmail to be 
rebuild, something there went wrong (I think).
When you do a "make setup check" all files in /var/qmail/ get overwritten. 
Do you keep your QMail files there? If not, try to manually build and 
install the binaries.

Else, give us some more info of what you tried.
