Did you restart qmail?

Does vmail meet the requirements in "man qmail-getpw" to be a valid
qmail user?

That much looks OK.

There are multiple "right" ways to do this. What happens if you do:
cp ~vmail/.qmail-domain1-user1 ~alias/.qmail-vmail-domain1-user1
?
