What does  qmail-qstat  say?
For quick queue inspection I use:

    | tr '[A-Z]' '[a-z]'        \
    | sort -t@ +1               \

This cummulates and counts the emails in the queue for each domain.
You may want to check the IPs of the MX for these domains against
qmail-tcpto.
