That's not specific to maildirmake: *all* commands have to be:
1) specified with an absolute pathname (/var/qmail/bin/maildirmake),
2) specified with a relative pathname (./maildirmake), or
3) in a directory contained in your "path".
This is *very* basic UNIX stuff.

If you installed using the LWQ directrions, you can do:
echo ./Maildir/ >/var/qmail/control/defaultdelivery
qmail restart
Otherwise, look at /var/qmail/rc or however/wherever you're running
qmail-start.
