Examine

    man -M /var/qmail/man qmail-users

It gives detailed instructions of the format of the assign file. Two
important things to remember

    1) the file MUST end with a single dot on a line by itself
    2) you MUST run /var/qmail/bin/qmail-newu for your changes to take effect
