The score to delete is "12" in my mailfilter. The marking of
"***SPAM***" is done by Spamassassin itself, not mailfilter.

The email never came through to the
client (thunderbird), so I am assuming Spamassasin is working, also I
checked the tail of the log and it showed that the message was treated
as spam. What happened to this email, where did it go? How do I know
what emails have been blocked/marked as spam, does a list get
populated or do I just look in the log manually.
You can check /etc/mail/mailfilter
Any spam scores above 21 will be deleted, and > 5 < 21 will be marked as ***
