You might accomplish this with file permissions and ownerships. Let the MDA have write access to the spools, but only let qpopper have read access. Not sure if that'll break qpopper, though. For instance, create the spool files with ownership root.user (ie. owner root, group set to the user for that spool file), and set the permissions to 740.
