It depends on the hub.  A standard port connects to a computer or 
printer via a straight through cable.  Many hubs and switches have an 
uplink port that is used to connect to another hub or switch.  It is 
connected to a standard port on the other hub or switch via a 
straight through cable.  It doesn't matter which device is used for 
the uplink port so it could be that what you perceive as an upper 
level hub might use it's uplink port to connect to a lower level 
device.  "Uplink" is a convenient term but not entirely accurate. 
The uplink port on a device is usually a standard port that is wired 
like a crossover cable so a straight cable may be used.

The standard ports on a hub/switch are known as MDI and uplink ports 
are MDI-X.  You can think of computers, printers and such as having 
MDI-X ports.  You connect an MDI port to a MDI-X port via a straight 
through cable.  You use a crossover cable to connect two MDI ports 
together or to connect two MDI-X ports together.

Each port (of either kind) has a pair of wires that is transmitting 
data and one for receiving data.  The transmit pair needs to connect 
to the receiving pair on the other device and vice-versa.

Some newer NICs do auto-sensing of transmit / receive.
