The locations themselves need to have what is known as a Wide Area Network or VPN for R:Synchronizer to work.
What R:Synchronizer does is run on a machine at location A and connect to a database there and a remote database at the same time, then based on rules you've established, synchronize data between the two databases. For this to work the system on which R:Synchronizer runs has to be able to see the remote database as if it were on the same local area network.
