Having the label aligned with the valve hole also helps you find whatever was stuck in your tire that gave you a flat. Once you find the hole in the tube, go to that position on the tire to look for the offending item. If the hole is 90 degrees from the valve, look at both places 90 degrees from the label on the tire.
