If you have been riding it all along with another wheel in there and it's been perfect, and this "new" wheel in question only is off center, then clearly that "new" wheel in question is the only issue here.
If you have not been riding the bike before and acquired it, then by all means state that as then the frame could be an issue, I say "could" Since we the reader know nothing of the history prior to you posting this, we are getting a very limited picture.
