I wouldn't do that as the preferences file is usually checked both at startup and quitting; creating a new one if it doesn't exist. If you delete it during running it will simply re-create the last one from memory. If you do it with Rb *not* running, it will create a new default one.

This is in 2005 and up. The clue will be you will see the EULA on startup.

In 5.5.5 you *can* delete the REALbasic Recent Items folder and it will stay empty until you open a project or save a new one.
