I'm pretty sure it is, as I've seen a cached reference to just such a
thing (Google for 'up2date server howto'), but I can't find a reference
as to what you'll need to do to actually set the server up. 
Client-side, you simply modify the settings in
/etc/sysconfig/rhn/up2date.

A nice alternative is to use apt-get via the apt-rpm project.  It is
definitely documented, including a security mechanism via gpg keys.  The
only problem is that you'll have to install apt on your 500 clients.
