What exactly are you trying to prevent?
****Do not do this!****
You can disable the root account by replacing the password
in /etc/shadow with "!!". Then no one can login as root or
use "su" to switch to root.

You can disable root logins on specific terminals by editing
/etc/securetty. That file is a list of terminals where root is allowed to
login.
You can change the root password and no give it to the user that you
want to restrict.
You can use sudo and give specific superuser privileges to specific
users.
