It will usually program in the Ham band but you will need to realign the receiver front end and the transmitter PLL. You will need the help of someone with a programming interface and software to program and with a service monitor and the book to align. The programming code has to be changed to work in the ham bands. The code is available on the internet as is the interface cable.
 
You may want to add an IDer since the R100 doesn't have one. Also, they came in several versions with differing output power, squelch (carrier, PL, or DPL), and control functions.

This is my first entry into repeaters, so I'm glad that I found this
group. 
A friend has offered me a Motorola R100 repeater (UHF) and duplexer if
I can figure out how to make it work as an amateur repeater.  From
what little I've learned so far, it appears that this will be an
out-of-band operation for this repeater, so more programming will be
required.  Right now we cannot find the key to the repeater chassis.
Can reprogramming be done without opening the case?
I've chosen a couple of promising frequency pairs and have been
monitoring them for traffic.

I'm hoping for advice and resources that will help me to get started
on this project.
