If your duplexer provides enough isolation to give you desense-free
operation when using a single antenna, then for sure it will be fine when
"split" to two antennas since you're only gaining additional spatial
isolation at that point.  But, if your duplexer was marginal to begin with,
then you would want to shoot for enough make-up spatial isolation as
necessary.

Using 100 dB as your target isolation (probably a fair number to use), as
long as your duplexer was capable of providing 70 dB or more of isolation on
its own, just separating the antennas vertically, and colinearly, by only
25' will easily push you above the 100 dB mark.
