In one variable

sage: x.subs(x==10) 

works ok. 

How to do a pair substitution ?

sage: var("x,y")
(x, y)
sage: (x,y).subs([x==10,y==20])
---------------------------------------------------------------------------
AttributeError: 'tuple' object has no attribute 'subs'
---------------------------------------------------------------------------
