I am not too familiar with xsane but as far as I can determine it completely 
supports all available functions.
I scanned several documents in color and lineart mode with no problems.
I used different resolutions, adjusted gamma, adjusted brighness, adjusted 
contrast, and got expected results.
The automatic adjustment of threshold in lineart mode worked properly.
The preview function worked properly.
'Toggle lamp of flatbed' under xsane 'Windows-show advanced options' worked 
properly.
I was a bit surprised at the xsane 'Toggle lamp of flatbed' function since the 
Microtek ScanWizard Pro 3.09 software and drivers for the Microsoft Windows 
operating system do not provide any functions for turning off the lamp.
The lamp never shuts off while the scanner is turned on when connected to a 
computer running Microsoft Windows.

I downloaded the 'live' CD of the 20090323 build of Ubuntu 9.04 alpha for 
64-bit computer systems to see if xsane 0.996 worked identically under Linux 
for 64-bit systems as it does under Linux for 32-bit systems and it does.

I did not make any configuration file changes; after booting the 'live CD' I 
started xsane and xsane detected the Microtek ScanMaker 6400XL scanner 
automatically and functioned perfectly.
I do not know if configuration file changes are even possible when running a 
'live CD' installation, where the operating system and applications are all 
loaded to RAM and nothing is saved to hard disk drive.

The 'info' function under xsane confirmed the loaded backend was microtek2, the 
device was /dev/sg2, and the sane version was 1.0.19.

I ran sane-find-scanner -v -v and the relevant output was:

how would you characterize the support- minimal, basic, good,
complete? and, just to be clear, it is the microtek2 backend which
supports the machine? did you have to make any configuration file

The Microtek ScanMaker 6400XL scanner is not listed at 
http://www.sane-project.org/sane-mfgs.html, either as supported or unsupported.
I was going to add an entry for the Microtek ScanMaker 6400XL as supported on 
the list at http://www.sane-project.org/sane-mfgs.html.
I only saw a method for adding unsupported scanners at 
http://www.meier-geinitz.de/tinc?key=rDoQ7lrj&formname=adddev.
Is there some method for adding supported scanners to the list?
