I could not find any formula. I think that there will always be a need for
trial and error. I would start with the recommended my.cnf for various DB
sizes. In one of the distributions of MySql I found a recommended my.cnf for
small, medium and large databases when the size is based on the machine's
memory.

If you can't find it I can take a look and try to find it for you....
