focus on cryo-electron tomography. The scientist will use cryo-electron

open-minded for new ideas

SwissFEl is now operating, which opens very unique opportunities for time 
resolved room temperature crystallography. We developed several membrane 
protein projects that need crystallography support. An interest in chemical 
mechanism, kinetics  and a good background in biophysics and crystallography

Within the Electron Microscopy Group (EMF) we have a strong user community for 
biology research projects taking advantage of the availability of our two 
transmission electron microscopes. The main focus is cryo tomography, single 
particle analysis and electron diffraction. Our high-end 200 keV TEM equipped 
with two direct electron detectors allows high-end data acquisition and the 
development of new techniques. For this group we are looking for a

Staff Scientist
Your tasks
As a part of a highly skilled and motivated team you will be involved in 
maintaining and further developing hard- and software in collaboration with PSI 
groups working in biology and development, in user training as well as 
implementing new techniques at the facility.


  *   Technical support for the 2 transmission electron microscopes at the EMF 
including daily maintenance
  *   Introductory trainings to new users at the EMF
  *   Advise and support scientists (mainly withing biological context) with 
their experiments at the EMF
  *   Further development of existing and implementation of novel technologies 
and procedures for the analysis and interpretation of cryo-electron microscopy 
data of biological specimens
  *   Expert support in sample preparation of biological specimens using the 
high pressure freezer and vitrobot at the facility
  *   Close collaboration with the manager of the facility for maintaining 
hard- and software and establishing new techniques at the facility
  *   Supporting and advancing important projects at the facility: high end 
single particle analysis, electron diffraction methods and electron ptychography

Your profile

  *   University degree at PhD level or equivalent in natural sciences
  *   Strong background and hands-on experience in electron microscopy is 
required
  *   Experience in cryo EM technologies (hardware and software)
  *   Strong interest in biological projects using electron imaging methods
  *   Strong communication and collaboration skills in an international 
environment
  *   Fluent communication in English
  *   Previous experience in user support and/or EM user facility is a strong 
plus

We are seeking a highly motivated and skilled PhD student for a protein 
engineering / structural biology project at the Paul Scherrer Institute (PSI) 
in Villigen, Switzerland. The aim of the project will be to engineer membrane 
proteins and membrane protein complexes that are suited for analysis by 
cryo-electron microscopy (cryo-EM) from single particles. The project will 
involve molecular cloning, protein expression in E. coli and mammalian cells, 
protein purification, biophysical characterization and analysis by electron 
microscopy. Ideally, the student should have a strong interest in molecular and 
structural biology and a strong interest in the newest structural biology 
methods. Experience in electron microscopy, protein expression in E. coli and 
mammalian cells, molecular cloning and protein purification would be highly 
beneficial. The project will be supervised by Dr. Roger Benoit and 
co-supervised by ETH Prof. Dr. Gebhard Schertler.

A postdoctoral position is available in the group of Prof. Volodymyr M. Korkhov 
at Paul Scherrer Institute (PSI) and ETH Zurich, to work on structure and 
function of membrane-associated signal transduction complexes using electron 
cryo-microscopy (cryo-EM) and single particle analysis.
The successful candidate will have expertise and a track record in cryo-EM and 
single particle analysis. The candidate will be experienced in molecular 
biology, including cloning, heterologous protein expression and purification. 
Previous experience with biochemistry of membrane proteins will be an advantage 
but not a prerequisite. The candidate will be able to work independently and 
will have excellent teamwork and communication skills.
The Korkhov group is integrated into the Laboratory of Biomolecular Research at 
PSI and has full access to all facilities and instruments for protein 
expression, purification, biochemical and biophysical analysis. In addition, 
the group benefits access to the PSI crystallization facility and the Swiss 
Light Source synchrotron radiation facility. The group benefits from the 
in-house 200 kV screening transmission electron microscope (JEOL JEM2200), 
planned to be upgraded with a direct electron detector. Furthermore, the group 
has full access to all research facilities at ETH Zurich, including the K2 and 
Falcon II direct detector-equipped Titan Krios microscope at the ETH Scientific 
Centre for Optical and Electron Microscopy (ScopeM).

The Laboratory of Biomolecular Research (LBR) 
(http://www.psi.ch/lbr/laboratory-of-biomolecular-research) in the Biology and 
Chemistry department of the Paul Scherrer Institute counts around 80 people 
pursuing fundamental research in large part in the field of transmembrane 
signalling. We work on membrane proteins that are essential for many biological 
processes and therefore are the targets of more than 30% of the drugs on the 
market. Research at LBR is therefore strongly linked to human health trough 
collaborations with academic laboratories and pharmaceutical industries.
With the very close proximity of the Swiss Light Source (SLS) synchrotron, the 
development of projects for studies at the swiss Free Electron Laser under 
construction (SwissFEL) and its excellent infrastructure, the LBR is an 
attractive environment for technical employees, PhD students and postdocs 
interested in structure/function analysis of complex biological systems.

Duties  Responsibilities:
-Dedicated support to the research projects in the group of Gebhard Schertler 
as well as maintenance of basic functions in the lab. Your main activity will 
be conducting experiments with protein production (PCR, plasmid preparation, 
bacterial and eukaryotic cell culture), protein isolation followed by 
biochemical characterisation (chromatography techniques, SDS-PAGE, Western 
blotting, stability tests and ligand binding assays) and crystallisation 
(manual and robotic setting up). You will carry out some experiments in 
low-light conditions in a photolab and occasionally give assistance during 
diffraction measurements of crystal samples at the SLS synchrotron which 
require some flexibility in the working hours.
-A part of the job (20%) will support the general LBR infrastructure and 
laboratory workflow both in an independent manner and in collaboration with the 
group.

Requirements:
As an ideal candidate, you must have a minimum education as technical assistant 
with some experience in biochemistry. You should have knowledge in protein 
production and purification and be highly motivated in working on fundamental 
research with protein and DNA. You are able to work as part of a team and 
independently with excellent communication in English. Keeping an electronic 
labbook as well as organisational (inventories) and basic administrative skills 
are essential.
A bachelor degree in biochemistry or chemistry, German knowledge, experience in 
cell culture, protein purification and/or crystallization is an advantage.
