Is it possible to set an Out of Office Reply from Exchange Server, or must I
do it from outlook, and manually connect to each mailbox I want to enable it
on and key in the msg?

Search for OOO* in the subject lines, in the archives of this list,
specifically targetting the last half of December. This has been asked
and answered a few times in the last week.

I'd cut paste it for you, but I know you've been subscribed to this
list for months now. :)

You can do it from an Exchange Server using an Outlook client installed
thereon or through OWA.
