I suggest an extended prime to ensure no air in the line then consider if more 
drops of xylene are needed on the slides.

Isn't the Prisma a Sakura product? Have you called the vendor to ask for 
technical support over the phone? They are usually good about doing this, even 
if you don't have a service contract. You should also mention the lot number of 
the tape that you're using so they can document and track the problem if it has 
happened to other users.

Hi Brittany,
Personally I do not like the HM525NX.  It is awkward and difficult to control 
with no manual advance except the control panel.  When advancing /trimming you 
are never certain how far forward the block will travel.  The UV light fitting 
in the lid always seems to get in the way of my field of view.  Ours doesn't 
have temperature control of the block holder which can make sectioning more 
difficult.

Looking at the specs of the cryostar I would actually opt for the NX70.

Hello everyone!

I am new to the Histology world. You all may know my previous coworker,
Rhonda Ford. She officially retired and I am so happy for her! With that
being said, I am in a pickle because the cryostat we had completely broke
down. We were having issues with it during my training period, so I never
officially got to use it. We are now looking for a new cryostat for our
department. My supervisor is asking me about different models and features,
but I don't know much about any of this. So I was hoping you all could
help! Here are the details:

We are looking at two different models:
- HM525NX
- CRYOSTAR NX50

We are also getting questions about UV versus fog mist (More specifically
the "Cold D" fog mist). So my questions are:

1) Which model do you think is a better investment?
2) Is it ok to buy used?
- It's about half the price, which my supervisor really appreciates!
- A new cryostat will have to come from overseas. The seller says used ones
will probably be available in the states.
3) Which is better, UV or fog mist?
4) Are there any other features that we should aim to include in our
purchase (e.g. height adjustment, vacutome, etc.)?

[Histonet] BenchMark ultra drawers

Greetings!
Earlier this month we had two slide drawers replaced.  Yesterday one of them 
opened during a run and would not close again.  Fortunately we were able to 
salvage the slide that was being stained in the drawer.  A little later I was 
removing the remaining few slides from the module and noticed wisps of smoke 
coming from the drawer.  I put the module in sleep mode and the drawer still 
would not close so I turned off the power.  As soon as I did this the drawer 
closed.

I phoned Roche and at their direction I turned the module on again and the 
drawer opened again immediately.  It would not go into ready mode so we tried 
operating the compressor manually but to no avail.  The drawer then made a 
crackling noise and smoke started coming out again.

Has anybody else had a similar experience???

Hi Jeanine,
- Film coverslips scratch very easily.  Any dust in the system will scratch the 
film.
- Staining is often compromised if the film coverslip needs to be removed.
- Specific storage conditions are required to validate the adhesive on the 
film.  If the storage conditions are not met the longevity of the tape is not 
guaranteed.

- Glass is better for photography
- Glass does not scratch as easily
- Less fussy with storage requirements
- Removal does not compromise staining quality.

Perhaps they did an orcein stain with H&E counterstain?  That would make the 
copper associated protein a dark red-brown colour but with other components 
looking like an H&E.

copper deposits stained in the routine H&E slides. Is anyone aware of how this 
could have been achieved? I assume it would either have to be an additive in 
the eosin or perhaps something naturally occurring in the tap water rinses?

Miller's stain is available from VWR.
I do not recommend the BSS to stain elastin - it is not strong enough and 
misses the fine fibres.

You can get Victoria Blue R (Colour Index 44040) from Sigma-Aldrich and
several others. I couldn't find anyone who offers a kit, only the dry dye.
There are other dyes called Victoria Blue, reported to give the same
results.

You'd have to find a method for preparing it as an elastic stain, and I
couldn't find such a method either with Google or in my old books. The
requester needs to supply you with a method. I suspect the requester is
reading an old article. There are stains for elastic tissue (which is I
suppose what you want to "exam thickness of artery") that are a lot easier
to get.

[Histonet] Slide Barcode Reader Fault on Ventana BenchMark Ultras

Greetings All!
We have 2 BenchMark Ultras that have been on-site for almost 2 years.  Since we 
transferred our routine work onto these modules we constantly get error 
messages pertaining to the slide barcode reader.  It often implies that there 
is a problem with the slide barcode.  Often, if it is transferred to another 
drawer it will read the barcode without problem and start the run.  Other times 
we give up and go home and when we return in the morning the module will read 
the barcode without issue.  Sometimes just moving the slide to the other module 
does the trick.

It is not uncommon for one run to go through without problem but when we try to 
add another it has a tantrum and won't read most of the slide barcodes.  
Changing the barcode itself often makes the situation worse.

Add in the problems with faulty dispensers and we are growing to dread having 
to use these machines.

Has anybody else had these problems?  Does anyone have any possible solutions?
