Hi Nancy,

If someone is certification eligible, we hire them into the same position (for 
us it's a C2 level Histotechnologist) and they have one year to get certified. 
The pay would be the bottom off the range, with a raise when they obtain their 
certification.  If they don't have the requisite experience, we hire them in as 
a C1.

Well, I would worry much less about over-fixation than I would under-fixed 
tissues.  Do you not have the option to have a weekend processing schedule that 
runs RT formalin?

Is your volume sufficient you could do an analysis of weekend vs non-weekend 
ER, PR, HER2 rates and see if they are statistically different?  You should be 
keeping the overall statistics already.  If they are not statistically 
different, that should ease your mind,

to do because we can readily gross duplicate sections. Obviously we can't adopt 
this approach for smaller samples because they're entirely submitted.  I have a 
game plan in mind, but would love some additional input! How'd you do it?

provide certification that we will properly dispose of and be responsible for 
the blocks in our possession if our research partners find them of no use. For 
some specific positive tissues, a single block blocks can be worth over a

It depends on who your most stringent governing body is.  If you have a lab in 
NY, or that takes specimens from NY, it's 20 years for both.  For CAP it's 10 
years.  There may be other states that have more stringent rules, etc.

[Histonet] slide printer

Hi All,

I am posting this for a friend.  Any info would be appreciated.

Any way we are looking at several slide printer options and wondered the 
following:

1)  What are you using?

2)  Did you first try any other vendors besides the one you are using now?

3)  Do you like what you are using now?

4)  Is it a one or two slide hopper - ie can you load and then select 
charged or non-charged slides?

5)  Are you using vendor specific recommended slides or have you 
substituted for a cheaper or other option?

6)  We are looking specifically at Leica, SlideMate and ESPO - do you have 
any experience of info from other users you know about these vendors?

Any and all information you are able to share would be appreciated.

You will need to give it a try, in order to know for sure.

Water quality is EXTREMELY variable, from community to community, and even the 
pipes, etc., within in your building can influence the water quality.

The chemicals used by the local water district can also vary, based upon the 
season, and your geographical location.

We have always accessioned specimens with multiple parts as 1, 2, 3, etc. and 
the corresponding paraffin blocks as A, B, C, etc.  We are in the process of 
changing this to letters for the different specimen parts and numbers for the 
blocks.  For those of you who do it this way, have you encountered any 
problems?  For example, if the specimen arrives and the different parts are 
numbered, do you simply convert them to letters?  Thank you.

I have used this for IHC, as well.  It's great stuff!  You can also use other, 
very viscous mounting medias, but the time it takes to dry/soak are longer than 
with Mount-Quick!

Slide Repair

1.  Soak coverslip off of slide in xylene.
2.  Piece slide back together.
3.  Apply Mount-Quick to slide, covering tissue. Allow to dry overnight.
4.  Place slide in cold water in refrigerator overnight.
5.  Carefully peal the Mount-Quick off the slide.  The tissue should come 
up with the Mount-Quick, if it does not stop pealing up the Mount-Quick and 
place back into the water overnight again.
6.  Carefully trim excess Mount-Quick from areas where tissue is not 
present. Leave at least ¼ inch of Mount-Quick around the tissue
7.  On a new slide that is wet with water, place the removed Mount-Quick 
and tissue in the center of the slide (make sure the tissue is next to the 
slide), removing as many air bubbles from under the Mount-Quick as possible.
8.  Cover the slide with a wet piece of filter paper then place a blank 
slide on top.  If you have multiple slides to repair then they can be stacked.  
Apply weight to the top of this and let stand overnight.
9.  Soak the dry slide in Xylene until the Mount-Quick is removed and 
coverslip as usual.

For a few years I've been involved in a program called letters to a 
pre-scientist. The idea is to reach middle schoolers as they are being 
introduced to the sciences. They have pretty high goals at this time, they want 
to be doctors and astronauts and engineers but they are just starting to learn 
about these things.
You become a pen pal/mentor of sorts and write letters to a child and they will 
write back to you. Last year I was writing to a boy in the Chicago area and 
this year it was a girl in LA. I always write about what I do and how important 
it is and include pictures of things like brain cells, muscle, fungus, bacteria 
and pictures of my lab. I always pick up a copy of the NSH coloring book and 
send it to them and tell them what they need to study to be a histotech and 
other than a hospital, where they can find a job. Of course we also tell them 
about other things like our families, pets, vacations, etc. at the same time. 

It's just a small thing but it plants a seed.

While President of the NSH, Vinnie DellaSperanza started a career day function

  The following has to do with histology and STEM (science, technology,

 and computers there were a few projects having to do with medicine,

 immunohistochemistry pictures included.  At the end of the fair, we awarded 
 almost 1.8 MILLION dollars of scholarships and awards to grades 7-12 
 students.  Not only that, our top winners get an all-expenses paid trip to 
 present at the ISEF (Intel International Fair) with 1,700 students competing 
 from all 50 states and 70 countries.  Wherever you are in the US, you have a 
 state fair.

  I would advocate for some of you so interested at the national, state or 
 local levels to promote histology, by getting involved as mentors for middle 
 and high school students to science fairs; especially those that could lead 
 to histopathology or other related projects that could lead into Intel 
 affiliated fairs resulting in great benefit to the student and a spread of 
 the word of histology into both the STEM world and general population.

  I've mentored for 15 years.  It can be done.  Molecular histopathology, 
 personalized diagnostics and therapeutics, advances in immunohistochemistry, 
 current controversies about breast biopsy diagnosis, or other disease with 
 newer classifications, PCR and RTPCR in histology, modern-targeted 
 therapeutics like in melanoma or colo-rectal carcinoma, FISH, digital image 
 analysis software for you computer geeks and on and on; the list is nearly 
 limitless.  Especially if you are close to or can contact biotech companies 
 or educational institutions to find co-mentors for grades 7-12 there are 
 histology-related science project possibilities in terms of data collection 
 and the scientific method and project presentation are nearly unlimited now.

  Be a mentor for or engage a grade 7-12 student, with the help of another 
 mentor or organization, to think about

page 8.  I am still studying for the qualification.  I've been using the two

are some Journal of Histotechnology articles I'd like to get my hands on, but 
have not been able to yet.  I'm also curious about the other books.

We are located in Columbus, OH which is a wonderful city with a population of 
1.75 million. We are the 16th largest city in the US, without the traffic

The only time a skin is above an 88305 is when it is just the 'covering' on 
something that has soft tissue involvement to the point that it then becomes a 
soft tissue specimen, rather than a skin.

The position of Clinical Histology Laboratory Manager here at The Ohio State

average block count is about 765/day and we have about 18 techs in histology.

[Histonet] FFPE used for EM

Hi Everyone,

I hope it's a great Friday for you all.  

Our renal folks were recently attending a meeting, and they came back with
some partial information.  (I am hoping some of you might have some
additional information.)  It seems that one of the lecturers mentioned that
some paraffins actually seemed to make a difference in the quality of EM when
they had to take tissue from the paraffin blocks.  I had never heard of this.
I am aware of using Carson's modification of  formalin to yield better EM
results, but not anything to do with paraffin.

Can anyone help me out here?

Maybe there is something new, but we had our inspection last week, and the
customized checklist used for our inspection does not require 2 identifiers.
It states:

NOTE: An unambiguous system of specimen identification coupled with a
legible, sequential cassette
and slide labeling system that withstands reagents and stains are essential
to fulfill this requirement. The
number of blocks processed and the number of slides prepared for each case
must be recorded.

ANP.21100 Phase II N/A YES NO
Are blocks identified adequately?

NOTE: Each block of tissue must be identified by the entire accession number
assigned to the case and
by any descriptive letter(s)/number(s) added by the prosector during the
dissection. If additional blocks
are prepared later, all lists and logs must reflect these additions.
Identification number and
letter(s)/numbers(s) must be affixed to all blocks in a manner that remains
legible.

ANP.21150 Phase II N/A YES NO
Are slides identified permanently with adequate, legible information?

NOTE: Each slide must be identified by the entire accession number and
descriptive letters unique to the
block from which it is cut. Other appropriate identifiers should be included
as applicable (e.g., levels of
sectioning). Automated prelabeling systems are acceptable. Regardless of
whether the identifying
information is on the slide or on a label, the information must be indelible,

The laboratory director is responsible for ensuring that slides are

I think  you will find that CAP is asking if 2 patient identifiers are used
throughout the entire process.  We engrave the blocks, slides and a computer

I agree with Liz.  It seems with a bio major and chem minor, that should take
care of the coursework.  You could do the IU online program, but if you are
self-motivated, I would just study on my own, if I were you.  If you don't
pass the first time, then at least you would know how close you were, and
could decide to take the course, or just to study a little more.  If you can
pass the exam, why waste the time and money unnecessarily?  It sounds like
you probably have a great background and experience already!

You might be eligible to sit for the registry already either HT or HTL, you
just need to have the correct number of credits in biology and chemistry to
meet the requirements.  I would call ASCP to see if you have the
requirements, it seems to me that you have the experience needed you just
need to make sure you have the credits.  That's my opinion.  All you need is
one year on the job training with a BS or BA to sit for the HTL.  I think I'm
correct on this, but if I'm not we'll know soon enough from the replys.

I'm thinking about getting my HTL certification.  I've been a bench scientist
in both academia and biotech for 21 years, which has included pre-clinical
animal research and subsequent histological applications.
I've set up an histology lab and have a lot of IHC but only basic staining
(mostly HE), as well as loads of animal tissue processing and some human
tissues.  I have a liberal arts degree (an AB) with a biology major and a
chemistry minor.  We have both a DVM and a few MD's on site, but for
pre-clinical and clinical research consult.  I would appreciate any insight
on getting certified through an online program, how many hours is required,
and what kind of mentorship is necessary.

[Histonet] Workload value of whole mount embedding and cutting

I like both, but use the MX because they are cheaper, and they are, if not
quite as good as, certainly a much greater value than the AccuEdge blades (I
do think that on average I go through the MX a little faster, but they are
still cheaper!!).  I alsoo like the MB22 from ThermoFisher for cutting small,
thin sections, like renal bx.

I have always done the quench 1st if I do it manually.  If it is automated,
then unless you have a newer instrument with onboard retrieval, you don't
have a choice but to quench after the retrieval step (and may not have a
choice then depending upon the system and how open it is).

I'd say it doesn't matter which is 1st, except my inclination is always to do
the quench step 1st because the tissues seem a little less delicate before
retreival than after.

When doing immunoperoxidase staining on a tissue that requires antigen
retrieval, which do you do first - the antigen retrieval or the endogenous
peroxidase blocking?  Or does it matter?
