What version of Samba ? There was an issue with Excel files
on 3.0.0 that is fixed in the 3.0.1pre release code (available
as source from samba.org).

had a similar problem here
links in exel sheets caused lockup of exel (to slow - or not opening of 
the links)
it seemed to be an oplocks question
try with

locking = Yes
oplocks = Yes
level2 oplocks = Yes
posix locking = Yes
strict locking = No
